Erasca Wins FDA Fast Track for ERAS-0015 Pan-RAS Drug

The US Food and Drug Administration (FDA) has granted Fast Track Designation to Erasca’s ERAS-0015, an oral pan-RAS molecular glue under development for metastatic pancreatic adenocarcinoma. Announced on 27 August 2026, the designation accelerates development of a candidate positioned in one of oncology’s highest unmet-need indications, ahead of a planned Phase 3 pancreatic cancer trial and two potentially pivotal Phase 3 trials in lung cancer.

ERAS-0015 is designed to inhibit RAS signalling across multiple RAS mutations rather than targeting a single mutation, positioning it in the emerging pan-RAS molecular glue class ahead of established mutation-specific competitors such as Amgen’s Lumakras and Bristol Myers Squibb’s Krazati, both targeting KRAS G12C only. Preliminary data from the AURORAS-1 Phase 1 trial reported in July 2026 showed a 57% unconfirmed objective response rate at eight weeks in patients with second-line or later KRAS G12X pancreatic ductal adenocarcinoma receiving ERAS-0015 monotherapy at the 32 mg once-daily recommended expansion dose. All responding patients remained on treatment at data cutoff. Additional data from monotherapy expansion and combination cohorts, including panitumumab combination, are expected in H1 2027.

FTD provides more frequent FDA interactions and eligibility for accelerated approval, priority review and rolling review. Pancreatic ductal adenocarcinoma has a five-year survival rate below 15% and is characterised by KRAS mutations in over 90% of cases, driving intense biotech competition around pan-RAS approaches from Revolution Medicines (RMC-6236), Boundless Bio, Verastem and Frontier Medicines.

The commercial signal is a rapidly advancing pan-RAS drug category positioned to substantially disrupt KRAS G12C mutation-specific franchises and open substantial commercial territory in pancreatic and lung cancer. Erasca’s FTD momentum places it firmly in the leading pan-RAS competitive pack. Expect Revolution Medicines, Boundless Bio, Verastem, Frontier Medicines and established KRAS players Amgen and Bristol Myers Squibb to sharpen positioning through 2027.
