Astellas Licenses Audax DAX-044 IL-12 Fusion in $1.3bn Deal

29th October 2026

Astellas Pharma has licensed Audax Biosciences’ DAX-044 solid tumour-targeting IL-12 antibody-cytokine fusion therapy in a global immuno-oncology deal worth up to $1.3bn. Announced on 28 October 2026, the deal secures exclusive global rights to a preclinical asset built on Audax’s AVIKINE tumour-concentration platform and strengthens Astellas’ long-term oncology pipeline ahead of looming Xtandi patent expiry.

The deal pays Audax up to $285m in upfront and potential option exercise payments, up to $1.05bn in development and commercial milestones, and double-digit tiered royalties on global sales. Audax retains co-funding and co-promotion rights in Australia and New Zealand. DAX-044 uses Audax’s AVIKINE technology, designed to concentrate IL-12 immune-stimulating activity within the tumour microenvironment and limit the systemic toxicity that has historically constrained IL-12 therapeutic development. The asset is at the IND-enabling stage, with Astellas planning to initiate a Phase I study in 2027 and taking over global development, manufacturing and potential commercialisation. Audax has described DAX-044 as showing a best-in-class therapeutic index for an IL-12 agent in preclinical studies, though independent clinical validation is yet to come.

The deal reinforces Astellas’ pipeline positioning as Xtandi (enzalutamide), co-commercialised with Pfizer and generating approximately ¥960.8bn ($6bn) in 2025 sales, approaches patent expiry. It follows a recent $400m Astellas research collaboration with AI Proteins on de novo miniproteins, signalling an aggressive external innovation strategy. GlobalData projects the global immuno-oncology market will reach $186bn by 2031, with checkpoint modulators and T cell engagers continuing to drive category growth. The AVIKINE tumour-targeted IL-12 approach offers a mechanistically differentiated angle beyond those dominant modalities, with potential for substantial expansion across solid tumour indications if the therapeutic index observed in preclinical models translates clinically.

The commercial signal is a strategic Astellas pipeline reinforcement against Xtandi patent cliff risk, with IL-12 tumour-concentration offering a differentiated immuno-oncology mechanism beyond saturated checkpoint and T cell engager categories. Phase I initiation in 2027 is the next key milestone. Expect sustained big pharma licensing activity in differentiated immuno-oncology mechanisms through 2027 and 2028.
