Typewriter Raises $56m for Non-Viral Gene Therapy Platform
Typewriter Therapeutics has emerged from stealth with a $56 million Series A funding round to progress its non-viral, re-dosable in vivo gene therapy platform toward the clinic. Announced on 3 September 2026 and led by RA Capital Management and AN Venture Partners, the funding advances Typewriter’s target-primed reverse transcription (TPRT) platform amid growing industry concern over safety risks associated with viral vector-based cell and gene therapy approaches.
The TPRT platform harnesses the R2 retrotransposon, a natural system that enables insertion of a therapeutic gene into a specific target site. The RNA-based system is delivered via lipid nanoparticle (LNP), avoiding the manufacturing complexity, cost and safety concerns increasingly associated with viral vector delivery. Typewriter’s initial development focus is on in vivo CAR-T therapies and hereditary liver diseases. The company plans to initiate non-human primate studies in late 2026 while establishing its first in vivo CAR-T development candidate. Re-dosability is positioned as a key differentiator, allowing repeat administration where clinical benefit warrants.
Typewriter debuts amid intensifying concerns over viral vector safety, including patient deaths across recent gene therapy trials from Sarepta, Pfizer, Intellia, Rocket and Capsida, and recent trial pauses from Novartis (lentiviral CAR-T after three immune-related patient deaths) and Bristol Myers Squibb (autoimmune trial due to transient inflammatory events). In vivo CAR-T is emerging as a strategic focus for major pharma seeking to simplify administration and address ex vivo manufacturing challenges, with recent high-profile deals from J&J (Sail Biomedicines), Eli Lilly ($2.4bn Orna Therapeutics acquisition), AstraZeneca and Sanofi. Typewriter joins a competitive field including Prime Medicine, Verve Therapeutics, Beam Therapeutics and Intellia.
The commercial signal is a rapidly consolidating gene therapy category where non-viral platforms and in vivo cell therapy approaches are emerging as strategic hedges against the safety and manufacturing challenges of viral vectors. Substantial venture capital validation is accelerating platform maturity. Expect Prime Medicine, Verve Therapeutics, Beam Therapeutics, Intellia and major pharma partners J&J, Eli Lilly and Sanofi to sharpen their positioning through 2027.
