In an exciting collaboration, researchers from Shimadzu Corporation and Vrije Universiteit Brussel have made strides in optimising extra-column band broadening (ECBB) within micro-flow capillary liquid chromatography (LC). Published recently, their work addresses ECBB challenges that can significantly impact chromatographic resolution. This advancement is set to enhance the precision of micro-LC, a critical technique in fields like pharmaceuticals and biotechnology.

The last few decades have witnessed significant improvements in high-performance liquid chromatography (HPLC), primarily driven by advances in column technology. Yet, as columns achieve superior resolution, the importance of overcoming ECBB in various instrument components becomes evident, particularly in micro-LC. This method, distinct from conventional LC, benefits from reduced inner diameter and fluid volumes, making it crucial to address ECBB efficiently. In this study, the researchers focused on ECBB contributions from different LC parts, using coumarin compounds for their experiments. Notably, they identified significant ECBB reductions with a standard micro-LC setup, highlighting the vulnerability of peptide analyses to post-column ECBB during gradient LC. This research is vital for developing reliable micro-LC systems that fully exploit column potential.
