Science Study: Psilocybin Prevents Chemo Neuropathy in Mice
A preclinical study published in Science on 6 September 2026 has demonstrated that prophylactic psilocybin prevents chemotherapy-induced peripheral neuropathy (CIPN) in mouse models and preserves mitochondrial trafficking in ex vivo human peripheral nerves. The study also demonstrates that a non-hallucinogenic 5-HT2A agonist (TBG) reproduces the neuroprotective effect, providing critical evidence for developing prophylactic strategies for one of the most common and disabling side effects of cancer chemotherapy.
CIPN affects up to 60% of patients receiving neurotoxic chemotherapy regimens including cisplatin, paclitaxel and docetaxel, with symptoms ranging from numbness and tingling to severe pain that can force dose reduction or premature treatment termination. No preventive therapies are currently approved. The study team, led by Heles et al., tested prophylactic psilocybin across mouse models with cisplatin, paclitaxel and docetaxel, including a six-cycle cisplatin regimen and a syngeneic oral cancer model. Two psilocybin pretreatments completely prevented cisplatin-induced mechanical hypersensitivity, with cycle-matched pretreatment maintaining protection across six chemotherapy cycles and more than eight months of follow-up, without measurable impact on tumour growth or systemic cytokine profiles in tumour-bearing mice.
Selective 5-HT2A antagonism with volinanserin completely abolished psilocybin’s neuroprotection, confirming the receptor-dependent mechanism. Significantly, the non-hallucinogenic 5-HT2A agonist tabernanthalog (TBG) reproduced comparable protection against mechanical hypersensitivity, suggesting hallucinogenic activity is not required for neuroprotection. Live-cell imaging in freshly harvested peripheral nerves from 29 surgical patients showed that cisplatin markedly impaired mitochondrial motility, while psilocybin preserved trafficking and transport velocity via the TrkB-Akt-PAK5-MAP2-KIF5B pathway. The findings position 5-HT2A-targeted prophylaxis as a candidate strategy that could substantially benefit both the psychedelic therapeutics category and cancer supportive care franchises.
The commercial signal is a substantial validation for both the psychedelic therapeutics category and non-hallucinogenic 5-HT2A analogue development. CIPN represents a major unmet need with substantial commercial potential in cancer supportive care.
